Chiamata a ragione "la Divina", Sarah Vaughan possedeva una delle più belle, profonde e modulate voci della storia del jazz. Con la sua tecnica originale ed elegante preferiva cantare su basi armoniche ritmiche e veloci e sui contrasti dissonanti tipici del Be Bop.

Sarah Vaughan si avvicina alla musica poco dopo aver imparato a camminare, quando all'età di tre anni comincia a prendere lezioni di piano. Impara presto e dimostra un ottimo orecchio musicale, che la faciliterà anche nel canto. Poco più che adolescente si esibisce come organista e solista del coro di una Chiesa battista della sua città.
Nel 1942 vince un concorso di canto all' "Apollo Theater" di Harlem, grazie alla sua splendida interpretazione della canzone Body and Soul. Tra il pubblico c'è il cantante Billy Eckstine, che sei mesi dopo la chiama per unirsi alla sua band. In tal modo Sarah perfezione l'uso della sua voce, e avrà l'opportunità di farsi accompagnare da talenti del calibro di Dizzy Gillespie e Charlie Parker.
Nel 1945 lascia la band e comincia una fortunata carriera da solista. Nel '47 sposa il trombettista George Treadwell, in seguito anche suo manager. La cantante si sposerà altre tre volte: col giocatore di football Clyde Atkins, da cui avrà un figlio, col ristoratore di Las Vegas, Marshall Fisher, e col trombettista Waymon Reed.
Alla fine degli anni '40 Sarah Vaughan incanta milioni di americani intonando soavemente brani come Tenderly e It's Magic, e nel decennio seguente sfornerà un successo dietro l'altro, con canzoni come Misty e Broken-Hearted Melody. La sua profonda carica interpretativa e l'incisivo "vibrato" della sua voce rendono divina anche la canzone più mediocre.
Negli anni anni '50 si eisbisce con i più grandi musicisti jazz del periodo, come Miles Davis e Jimmy Jones, ed incide con successo una serie di dischi per le etichette "Mercury Records" e "EmArcy Records". Negli anni seguenti il successo continua e la sua voce, che man mano acquista un tono più baritonale, continua ad incantare le platee. Negli anni '60 affronta con successo anche il genere della Bossa nova.
Nel 1981 riceve un premio Emmy per un suo special televisivo dedicato a George Gershwin, e nel '83 le viene consegnato un Grammy alla carriera.
La "divina" continuerà ad esibirsi fino a poco tempo prima prima della sua morte, avvenuta all'età di sessantasei anni.
